A robust method for producing human mesencephalic DA neurons from hiPSCs is described and it is demonstrated that expanded mFPPs have increased DA neuron potential and differentiate more efficiently and rapidly than progenitors generated by standard protocols.

It is demonstrated that exogenous basic fibroblast growth factor (bFGF) could be omitted from the hPSC culture media if the authors used the suitable feeder cells, and it was shown that although hPSCs can proliferate on the feeder-free culture system, however, genetic instability has been reported in such environment.

The data indicate that cell line‐specific differences dominate the variables that underline the differentiation efficiency in the early stages of differentiation, and EB generation method and maintenance conditions determine the later differentiation and maturation of retinal organoids.

The results demonstrate that the extracellular matrix-modified PAA-ACA gels support mouse and human neuronal cell attachment throughout the differentiation and maturation stages (14 and 28 days, respectively) and emphasize the utility of optimizing numerous biophysical cues for improved neuronal yield in vitro.

The study implies that the numbers of prostate cells with stem/progenitor activity may be underestimated based on currently employed assays, supports that dissociation-induced apoptosis is a common feature of embryonic and somatic stem cells with an epithelial phenotype, and highlights the significance of environmental cues for the maintenance of stem cells.

Application of a selective Rho-associated kinase (ROCK) inhibitor, Y-27632, to hES cells markedly diminishes dissociation-induced apoptosis, increases cloning efficiency and facilitates subcloning after gene transfer, and enables SFEB-cultured hES Cells to survive and differentiate into Bf1+ cortical and basal telencephalic progenitors.

Results indicate that inhibition of Rho/ROCK signaling may improve survival of grafted cells in cell replacement therapy and suggest that dissociation of neural precursor cells elicits an intrinsic pathway of cell death that is at least partially mediated through the Rho-ROCK pathway.
